New clarification added: Dear bidders,Please find below clarifications/ responses given to questions received from ptential bidders:Question 1What parameters should be assessed during the method validation? As a supplier we are unable to validate a method as method validation is based on laboratory SOP's. Is a method verification acceptable?Clarification/ Response:Yes, method verification is acceptable. The method validation parameters required by end users include, but are not limited to, accuracy, precision, specificity, sensitivity, linearity, range, robustness, carryover, calibration curve, system suitability (retention time, peak shape, and resolution), matrix effects, and stability. We recognize that method validation is governed by the laboratory's specific Standard Operating Procedures (SOPs). While method verification is acceptable, we aim to ensure that the instrument can demonstrate and consistently reproduce or meet the pre-established performance characteristics of previously validated methods.​Question 2Please specify which analytes must be analysed in each sample matrix. Also specify what bioanalytical matrix is required (blood, urine, dried bloos spots etc.)Clarification/ Response: From the end users perspective, the analytes and matrices vary based on the type of laboratory requesting service to get from this instrument. Common analytes in bioanalytical applications include drug metabolites, biomarkers, and active pharmaceutical ingredients (APIs), while pharmaceutical wing of the institute may bring analytes involving APIs, excipients, and finished pharmaceutical products. Matrices generally include blood (plasma or serum), urine, and possibly dried blood spots, as well as various pharmaceutical formulations (e.g., tablets, capsules, or suspensions). The specific analytes and matrices to be analyzed will be determined based on the analysis  requirements. ​Question 3One of the specifications indicates that ESCi source is required, this is a lockout specification for a certain manufacturer. Please adjust accordingly. Clarification/ Response:Requesting ESI as a mandatory requirement is not a lockout specification. Not really. End user requested ESI as a must but also includes APCI ion source. End user has bioequivalanece, bioanalytical and pharmaceutical laboratories. As you are aware, In LC-MS/MS system, ESI (Electrospray Ionization) is ideal for bioanalytical applications, offering versatility and sensitivity for polar and thermally labile compounds, including small molecules, peptides, and drugs in biological matrices like blood, plasma, and urine. APCI (Atmospheric Pressure Chemical Ionization) is suitable for non-polar compounds and is effective in pharmaceutical quality control, especially for lipophilic drugs and formulations. If high-resolution and accurate mass measurements are needed, especially for unknown compounds or metabolomics, qTOF (Quadrupole Time-of-Flight) provides high resolution and accurate mass analysis. MALDI (Matrix-Assisted Laser Desorption/Ionization) is best for larger biomolecules, but it's less common for routine pharmaceutical or bioanalytical analysis. Overall, since we are purchasing three LC-MS/MS instruments, two of them should include ESI and APCI ion source in the same LC-MS/MS platform whereas the third LC-MS/MS has to be qTOF ion source.Bidders are advised to note that this ITB will be amended to reflect this change in quantity and a fresh ITB will be publishe separately for the requirement for LC-MS/MS with qTOF ion Source . For our purpose, we don’t need FAB, DESI and MALDI (Matrix-Assisted Laser Desorption/Ionization) ion sources.  If we requires high-resolution structural analysis of complex compounds (such as unknown metabolites, drug degradation products, or detailed pharmacokinetic studies), we will use our qTOF with better choice due to its superior resolution and mass accuracy. If we focus is more on quantitative bioanalysis, drug testing, and routine pharmaceutical QC, and we do not require the high resolution or structural elucidation capabilities of qTOF, and in that case we will use our ESI would likely be more appropriate. That is why we needed instruments with both ion source versionsQuestion 4:The Diode Array Detector specification requires spectral library/data base. This would not be required as a mass spectrometer is included in the configruation. Please adjust accordinglyClarification /Response:We appreciate your insight on this matter. As the LC-MS/MS system will serve both bioanalytical and pharmaceutical quality control applications, the mass spectrometer itself will provide the necessary spectral analysis and detection capabilities. Therefore, the requirement for a Diode Array Detector (DAD) with a spectral library/database can be excluded, as the mass spectrometer’s sensitivity and specificity are sufficient for the intended applications. The platform submission states that this form is mandatory to provide with the bid submission. However, the tender requirements also indicate that it should be provided within two weeks after the award.Response/ Clarifications:Bidders must fill in and submit Form F ''Performance Statement Form'' with their bid.
